In collaboration with the American College of Gastroenterology, this webinar featured Katharine Germansky, MDÂ from Beth Israel Deaconess Medical Center, and Pooja Dharwadkar, MDÂ Â from University of California, San Francisco, who delivered an in-depth look at
chemoprevention in hereditary cancer syndromes, focusing on Lynch syndrome and familial adenomatous polyposis (FAP).
Dr. Germansky began by reviewing the characteristics of an ideal chemopreventive agent and the challenges of identifying effective candidates. She highlighted evidence for aspirin, including its mechanism of action and findings from the CAPP2 trial, and discussed decision-making around its use. She also touched on other potential agents such as naproxen, statins, multivitamins, calcium, and the role of chemopreventive interventions in extracolonic cancers using resistant starch or hormonal therapy. She concluded with emerging data on GLP-1 agonists and their potential future role in Lynch syndrome.
Dr. Dharwadkar then shifted focus to FAP, providing an overview of the syndrome, associated cancer risks, and management. She reviewed the goals of chemoprevention in FAP, the mixed evidence for NSAIDs, specifically sulindac and COX-2 inhibitors, and investigational agents such as eflornithine, erlotinib, and mTOR inhibitors. She closed with an overview of current guideline recommendations.
This session offered valuable insights into current and emerging strategies in chemoprevention, and what may lie ahead.
